Next generation sequencing of mtDNA from putamen of young and aged Caucasian male donors showed that the frequencies of the common deletion, m.8483_13459del4977, as well as clonal and non-clonal deletions in the major arc between the mtDNA origins of replication, are significantly increased in the aged cohort (the fold increase in the frequency of the common deletion exceeding that of major arc deletions). Additionally, single nucleotide variants also increase with age with the highest rate of accumulation in the non-coding control region.
